Taxanes (Paclitaxel, Docilitaxel)

  • Taxanes act by stimulating polymerization of microtubules.
  • Paclitaxel was synthesized initially from the bark of Yew tree, but this source is limited. Hence now a days the precursor for paclitaxel is synthesized by genetic engineering in E.Coli.
  • It is used for treatment of breast, ovarian, genitourinary, lung and head and neck cancer.
  • Bone marrow suppression is a most frequently seen sideeffect and hence is routine combined with filgrastim to prevent neutropenia. Other sideffects seen are hypersensitivity, peripheral neuropathy and mucositis.
  • Vinka alkaloids (Vincristine, vinblastine, vinorelbine) act by inhibiting polymerization of microtubules.
  • Epothilones (Ixabepilone) acts by stabilizing microtubules.
