General Parasitology and Protozoa
In formol ether concentration technique, which layer contain the parasites? (AIIMS Nov 2009)
Concentration techniques include:
· Sedimentation techniques
a. Formol ether concentration technique
b. Formol detergent concentration technique
· Floatation techniques
a. Sodium chloride floatation techniques
b. Zinc sulphate floatation technique
c. Sugar floatation technique
Formol ether concentration technique is overall best sedimentation technique. It is rapid and gives good results.
Extra Edge: The zymodemes of the amoebae are studied by the mobility of 4 isoenzymes (L-malate: NADP + oxidoreductase; glucose phosphate isomerase; phosphoglucomutase and hexokinase). A total of 22 distinct zymodemes have been demonstrated in the amoebae population. The amoebae populations showing 7 (ii, vi, vii, xi, xii, xiii, xiv) zymodeme patterns are pathogenic while the amoebae showing other zymodemes are non-pathogenic. Pathogenic amoebae showing zymodeme xiv are exclusively found in India.
