Uterine relaxant with least side effects
Nifedepine. Studies have shown that the drug is equally effective compared to routinely used drugs in premature labour.
a. Ritodrine can cause side effects like palpitations, arrythmias, ischemia and pulmonary edemA. Recently, it has been realized that there is a need of developing new drugs to arrest progression of premature labour.
b. Though magnesium is better tolerated by magnesium can produce hypotension, palpitation, headache and muscle weakness.
c. Magnesium sulphate is still an off label drug for treatment of premature labour. Rarely cardiac arrest or respiratory failure can also occur.
d. Calcium channel blockers reduce calcium influx into cells, reducing muscle contractility. Nifedipine has been used in the majority of studies.
e. A 2002 meta-analysis reviewed 12 randomized-controlled studies involving 1029 women and found that nifedipine is more effective than ritodrine and is clearly safer.
