- 5-FU is a metabolite of flucytosine and inhibits the ‘thymidine synthetase’. It is a prodrug and requires intracellular conversion into the active and phosphorylated metabolites. It is a short acting drug with half life of 10-15 minutes. It is poorly absorbed by oral route and hence is best given by IV route.
- It is the drug of choice for colorectal cancer. Most common side effect is gastrointestinal upset. Other side effects include hand foot mouth disease and bone marrow suppression
