Lugol’s iodine test

Explanation

a. Lugol’s iodine test is done on the washing of glans penis (or from a moist blotting paper) of the accused which is then exposed to Iodine vapors or Lugol’s iodine solution. Brown color means the test is positive (results can be elicited up to 4th day), brown color is due to the presence of glycogen in the vaginal epithelial cells of the victim.

b. This test is an important tool in the examination of the accused person of a sexual offense/rape.

c. Presence/absence of smegma should also be noted.

d. Presence of smegma rules out complete penetration as it gets rubbed off during sexual intercourse.

e. It is collected again after 24 hrs.

f. A non-retractile prepuce is one which covers the glans penis.
